To make it work, you need to keep an efficient pace and, above all, carry the skip-the-line tickets booked for both monuments. A typical day would look like this:

The transfer: from the Vatican to the Colosseum by metro
By metro it takes about 30-40 minutes. From Ottaviano (line A, next to the Vatican) you go to Termini and change there to line B to the Colosseo stop, right opposite the Colosseum. Why it is a tight day
The Vatican Museums are huge (several kilometres of galleries) and the Colosseum, Forum and Palatine together also take time. Chaining them in one day means little rest and a lot of walking. It is feasible if you keep a good pace, carry skip-the-line tickets and start early, but you'll end up tired. Alternative: do it over two days
